Retatrutide (LY3437943) is a GIP/GLP-1/glucagon triple receptor co-agonist in Phase 3 clinical investigation. Phase 2 NEJM data (2023) demonstrated up to 24.2% mean body weight reduction at 48 weeks in the highest-dose arm, versus 2.1% with placebo.

Retatrutide (LY3437943) is a single peptide that activates three metabolic receptors: GLP-1, GIP and glucagon. The first-generation incretin compounds act on one of these (semaglutide on GLP-1) or two (tirzepatide on GLP-1 and GIP). The added glucagon-receptor activity is the reason retatrutide is studied separately from them: in published research it is associated with higher energy expenditure and hepatic fat oxidation, not only reduced intake.
Receptor-binding work reports the strongest potency at the GIP receptor with balanced activity at the GLP-1 and glucagon receptors, and a 2024 structural study in Cell Discovery describes how one conserved core sequence engages all three receptors. The most cited dataset is the Phase 2 obesity trial published in the New England Journal of Medicine (Jastreboff et al., 2023), which reported a mean 24.2% reduction in body weight at 48 weeks in the highest-dose arm. Eli Lilly's TRIUMPH Phase 3 programme is ongoing, and retatrutide is not approved for any use in any market.
